Short-term and long-term instrument reliability and robustness are critical issues in bead dispensing. Beads have the potential to become lodged in valves, reducing the lifetime and increasing the downtime of many traditional liquid handlers. Innovadyne’s proven valve-free flow path technology isolates the syringes and solenoid valves so that they remain in a pristine system fluid environment (degassed, de-ionized water) and the beads never come in contact with moving parts.

To address bead suspension a peripheral employing a paddle-wheel stirrer in the reagent tray, or an orbital shaker beneath the source plate can be used. Additionally, a tip harness, customized for the application, has been developed that keeps the dispense tubes vertical, facilitating even distribution of the beads. A protocol is put in place that optimizes the aspiration rate and dispenses beads rapidly (~10 seconds for a 384-well plate and 15 seconds for a 1536-well plate) before they have a chance to settle in the dispense tubing.

Traditional and flow-through liquid handlers can be destructive to live cells, and create problems with contamination and clogging of flow paths. In contrast, Innovadyne instruments have a valve-free fluid path and can perform fast dispensing of cells without the stress that occurs with flow-through solenoid or peristaltic technologies. The Nanodrop has dispensed sensitive cell lines, such as MV 411 Leukemia, with no loss of viability. Subsequent dosing of cell assays can be performed over a dynamic range of five orders of magnitude, with volumes of 50 nL to 500 µL.

The result is significantly improved cell viability, increased throughput for sample processing, and reduced volume of reagents. Innovadyne’s easily changed nozzle harnesses also facilitate sterilization protocols.

Innovadyne produces high-precision, non-contact dispensing instruments capable of reducing total-assay test volumes by at least an order of magnitude. The Nanodrop™ series’ highly precise, fast, non-contact reagent dispense not only reduces traditional assay costs tenfold, but also improves signal-to-noise and data quality.

Non-contact dispensing can also aid in mixing and in the elimination of trapped air in the wells, improving detection. The Innovadyne aspirate and dispense architecture — with a valve-free fluid path — has enabled robust assay performance even with problematic reagents. Chemistry dependent washes can be executed with intermediate solvents followed by a DI water flush. To perform MALDI-TOF mass spectrometry, compounds must be spotted on a flat, metal MALDI target plate. Positional accuracy and precise deposition of the spots is critical. Classical liquid handlers using contact methods are prone to failure due to plate and tip irregularities, poor positional accuracy (typically > 100 µm/axis) and drop morphology (doughnut effect ). Innovadyne’s platforms reliably fulfill critical MALDI spotting requirements with precisely manufactured motion control (<20 µm return to spot accuracy), and high-precision, non-contact solenoid dispensing. The aspirate/dispense capabilities of the platform enable rigorous washing to be performed between samples.

The Nanodrop II is an ideal benchtop solution for use in spotting MALDI plates for protein characterization and quality control of synthetic oligos, as well as analysis of laser capture microdissection (LCM) cells. It includes two plate nests for source and MALDI plates. Single tip dispensing can be performed for rare compounds, and independent channel spotting enables spotting of unique plate maps.

Innovadyne liquid handlers help miniaturize PCR reactions (5 to 10 fold reduction in total reaction volumes) without compromising data quality. Decreases in both reagent and sample consumption lower costs and increase scalability, further enabling genome-wide approaches.

Innovadyne’s protein crystallography dispensing platforms deliver high-throughput crystallization screens and protein addition for sitting drop, hanging drop, and microbatch protocols. The cost of producing purified protein for high-throughput crystallization screening is expensive, and requires the use of robots that can dispense the protein without much waste and without denaturation. The low volume, non-contact dispensing and fast cycle times of high-throughput protein crystallography are an excellent fit for Innovadyne’s technology.

The Screenmaker 96+8 is our top-of-the-line, fully automated platform for protein crystallography experiments. It is the fastest platform on the market for crystallization screen performance – less than 45 seconds for a 96-well sitting drop screen. It can replace 3 liquid handling stations in one multi-faceted application: screen creation, screen set up and screen performance. The Screenmaker is also capable of preparing fine and coarse mother liquor screens/grids. The Nanodrop is a medium priced, semi-automated benchtop platform for performing crystallography screens and protein addition.
